Introduction Manual Overview This manual prov ides the user with all the information needed to install a NTE/CCD Detector and place it in operation. Topics covered include a detailed description of the NTE Detector, installation, microscopy applications, cleaning , specifications and more.

Chapter 1 General Information Figure 1. NT E/CCD Det ectors and Camer as Descriptio n The NTE/CCD air-cooled detector or cam era is ideally suited for m edium light lev el applications.
10 NTE/CCD Detector Manual Version 2.A The NTE/CCD detector is available in both C-m ount and F-mount config urations. See Chapter 5 for detailed information on microscopy.
Chapter 2 Detector Setup This chapter covers the setup procedures for both im aging and spectroscopic applications. General Instru ctions The following item s are applicable to both im aging and spectroscopic system s. Connecting the D etector Each detector is supplied with a cable to connect to the controller.
12 NTE/CCD Detector Manual Version 2.A suitable Gain setting. I n situations where the A/D rang e exceeds that of the array, it will generally be better to set the Gain to HI so that the signal can be spread over as m uch of the A/D rang e as possible.
Chapter 2 Detector Setup 13 Square-form at CCDs can also be used for spectroscopy, although they are m ore often used in imag ing applications. For square form at CCDs (e.g. 512 × 512 or 1300 × 1340), the user may orient the CCD to achieve binning along either direction of the CCD.
14 NTE/CCD Detector Manual Version 2.A Mounting Di rect i ons 1. Mount the flange to the detector using the two half-ring s and the screws provided. Note that the tapered side of each half- ring faces the adapter (Figure 5). 2. Next, thread the 10-32 hex screws halfway into three of the six tapped holes in the spectrograph’s exit plane.
Chapter 2 Detector Setup 15 Second Shutter Type Mount i ng Di rections The second shutter mount, used with all A cton Research spectrographs, requires no disassem bly.
16 NTE/CCD Detector Manual Version 2.A To Mount the Lens on t he Det ect or: 1. Locate the large indicator dot on the side of the lens. 2. Note the corresponding dot on the front side of the adapter. 3. Line up the dots and slide the lens into the adapter.
Chapter 3 Cooling the Detector Introduct ion The NTE/CCD detector is desig ned for cooled operation. A four- stage Peltier effect thermoelectric cooler, driv en by closed-loop proportional- control circuitry, cools the CCD. A thermal sensing diode attached to the cooling block of the detector monitors its temperature.
18 NTE/CCD Detector Manual Version 2.A 4. On the top side of the Temp knob is a rectang ular window that denotes hundreds of degrees C. Each com plete turn of the knob is - 100°C. Around the mov eable part of the knob are num bers from 0 to 99, in increments of 2.
Chapter 3 Cooling the D etector 19 Detector loses Temperature Lock The internal temperature of the detector is too hig h. This mig ht occur if the operating environm ent is particularly warm or if you are attempting to operate at a temp erature colder than the specified limit.

Chapter 4 Focusing Introduct ion Detectors for either im aging or spectroscopic applications must be focused for m aximum resolution. Im aging applications require adjustment of both the lens and the lens adapter. Spectroscopic applications demand both focusing and alignment of the detector to the spectrograph.
22 NTE/CCD Detector Manual Version 2.A fluorescent overhead lamps have good calibration lines as well. I f there are no “line” sources available, it is possible to use a broad band source, such as tungsten, for the alig nment. I f this is the case, use a wavelength setting of 0.
Chapter 4 Focusing 23 Throughput: The throughput of a lens is determined by its aperture, which can ordinarily be set to a num ber of different values, or f/ stops.
24 NTE/CCD Detector Manual Version 2.A Imaging Field of View When used for two-dim ensional i maging applicat ions, PI CCD detectors c losely imitate a standard 35 mm detector. Sinc e the CCD is not the same size as the fil m plane of a 35 mm detector, the field of view at a g iven distance is som ewhat different.
Chapter 4 Focusing 25 Remount the lens to the adapter and set the lens focus adjustment to the target distance. 4. Power up the system and, using the application software, select FreeRun , and Safe mode (Asynchronous). 5. Choose a fast exposure (0.1 s) and begin data collection by selecting Focus (WinView/32).

Chapter 5 Microscopy Applications Introduct ion This chapter discusses the setup and optimiz ation of your dig ital imag ing sy stem as applied to microscopy . Since scientific grade cooled CC D im aging system s are usually em ployed for low light level m i croscopy, the m aj or goal is to m aximiz e the light throug hput to the detector.
28 NTE/CCD Detector Manual Version 2.A Mounting the De tec tor on the Micros c ope The detector is connected to the microscope v ia a standard type m ount coupled to a microscope specific adapter piece. There are two basic detector- mounting desig ns, the C-m ount (standard) and the F-mount (optional).

Chapter 5 Microscopy Applications 31 Note: T he detector lens mount itself also has a focus adjustm ent. Although it is unlik ely that you would ev er need to use this adjustment in operation with a m icroscope (it is preferable that you use the relay -lens focus adjustm ent), it could be used if necessary.

Chapter 6 Operation Introduct ion Once the NTE/CCD Detector has been installed and its optics adjusted as explained in the preceding chapters, operation of the detector is basically straightforward.
34 NTE/CCD Detector Manual Version 2.A If y ou observe a sudden chang e in the baseline signal y ou may hav e excessive hum idity in the vacuum enclosure of the detector. T urn off the system and have the detector repumped before resum ing normal operation.
Chapter 6 Operation 35 Note that a cam era having the 35 mm shutter can only be used with an ST- 133 equipped with the 70 V shutter option. An ST-133 that has the 70 V shutter drive option can be identified by the 70 V OPT label on its rear panel as shown in Figure 14.
36 NTE/CCD Detector Manual Version 2.A Shutter Overheating The 25 mm remote-m ounted shutter for spectroscopy has a built-in therm al interlock to prevent ov erloading of its coil. I f run at a high repetition rate, the shutter m ay heat enough to trig ger the interlock , disabling the shutter.
Chapter 7 Cleaning Turn off all power to the equipment and secure all cov ers before cleaning the units. Otherwise, damag e to the equipment or injury to you could occur.
